Sodium Channels as Targets for Neurotoxins: Mode of Action and Interaction of Neurotoxins with Receptor Sites on Sodium Channels

Several excellent, comprehensive reviews have been published on the structure, physiological effects and binding properties of the various neurotoxins acting on voltage-sensitive sodium channels (e.g. Catterall. 1980; 1986; Strichanz et al., 1987; Baden, 1989; Norton, 1991; Martin-Eaudair and Couraud, 1995). This chapter is focused on the recent developments in the use of neurotoxins as molecular probes of sodium channel structure and function and cites original literature that has appeared subsequently to the previous reviews of this area. The aim of this presentation is to discuss, on the molecular level, some aspects of the interaction between several neurotoxins and their receptor sites, located on the sodium channel protein. The dynamic mutual modulation of the conformational, functional states of the sodium channel and ll1e binding of toxins will be accentuated.
